Endolysosomal network in B cell activation and immunity: Decoding novel signaling roles and antigen processing

B cell activation is the driving trigger towards mounting of antibody responses. In this project, we will tackle the previously unknown questions about the mechanism of early B cell activation initiated by recognition of specific antigens. We will examine the novel functions of the intracellular vesicle network, namely lysosomes and their regulation by septins, in the regulation of B cell activation. We aim to reveal molecular level insights to build better basic understanding of the cellular requirements behind immune responses. To reveal the therapeutic potential of inhibiting lysosomal activities in B cell pathologies, we examine the effects of targeting these pathways in clinical samples of B cell lymphoma or B cells isolated from the inflamed joints of patients with rheumatoid arthritis. Moreover, we will examine the viral antigen processing upon B cell activation to understand the molecular details behind successful vaccination responses.
