Design Principles for Protein-Nucleic Acid Hybrid Materials

RNA therapeutics need carriers that can protect fragile RNA molecules and release them in the right place at the right time. Current delivery systems are powerful but can suffer from structural and payload heterogeneity, complicating optimisation and reproducibility. This project aims to transform protein-DNA-RNA hybrid materials from empirical demonstrations into predictive, payload-independent carrier systems. It combines DNA origami with viral capsid proteins to create programmable nanoscale containers where RNA loading, protection and release can be optimised independently. Using advanced microscopy, biophysical measurements, and cell-free and cell-based assays, the project will determine how carrier structure, surface properties and stability affect RNA delivery and translation. The results will establish reusable design rules for safer, more reproducible RNA delivery platforms. The research is carried out at Aalto University with leading international collaborators.
